Welcome back, warriors! In this episode of Peptide of the Week, JD Denham and William T. Haas get back to basics and break down two compounds people have been asking about. ARA-290 for nerve pain and neuropathy, and 5-Amino-1MQ for fat burning and metabolic optimization. Two very different compounds with very different objectives.

We cover:

🧠 ARA-290 — Nerve Repair and Neuropathic Pain

– First discovered and researched in the early 2000s, still experimental but with human trials

– Selectively binds to the Tissue Protective Receptor (TPR) on immune cells, nerve cells, and metabolic tissues

– Reprograms macrophages from pro-inflammatory to anti-inflammatory, actually healing the nerve instead of masking pain

– Used for small fiber neuropathy, sarcoidosis, autoimmune nerve conditions, diabetic neuropathy, and post-viral recovery including long COVID

– Unlike painkillers, effects continue AFTER you stop taking it because it is healing the nerve, not just covering the pain

Real-world reports:

– Burning pain dropped from a 7 out of 10 to a 2 out of 10 within 3 weeks in diabetic neuropathy patients

– Mood lift and mental calm reported within 4 days (not the main intention but consistently reported)

– Nerve fiber density measurably increases over days to weeks

– Effects often persist 3 to 6 months post-cycle

What to stack with:

– BPC-157 and TB-500, NAD+, alpha lipoic acid, methylcobalamin, and B vitamins

What to avoid:

– NSAIDs and corticosteroids will blunt the effects

– Alcohol, high sugar, ultra-processed food, and chronic sleep deprivation all work against nerve healing

🔥 5-Amino-1MQ — The Fat Burning Compound

– Not a peptide, technically a small molecule compound. First developed around 2014

– Blocks NNMT (Nicotinamide N-Methyltransferase), which is massively upregulated in obese fat tissue and actively drives fat storage

– Blocking NNMT shifts your metabolism to burn fat instead of store it, essentially injecting keto without the diet

– Also replenishes NAD+ levels that have been depleted with age

– In animal studies: obese mice lost 7 to 9% of fat with no loss of lean muscle while eating the same amount of food

– Adds an additional 7 to 9% fat loss on top of GLP-1 results because it works on a completely different pathway

Who it is for:

– Anyone on a GLP-1 who has plateaued, this will relight the fire

– People with insulin resistance or pre-diabetes, it increases insulin sensitivity

– Already lean people looking to get even leaner and enhance body composition

– People with metabolic syndrome or NAD+ depletion

Best stacks: GLP-1 (especially R), MOTS-C, NMN, NAD+, L-Carnitine, Alpha Lipoic Acid, CoQ10
